PromoterExpressionLevel:ThisplasmidcontainsthemammalianCMVpromotertodrivegeneexpression.WehavetestedallofourmammalianpromotersinarangeofcelltypesandCMVisconsistentlythestrongestinthosewehavestudied.HowevertherearemanyreportsoftheCMVpromoterdemonstratingsilencingbymethylationinlong-termculture.ForthisreasonwestockarangeofotherpromotersthatarecompatIBLewiththisplasmidandareavailableonrequest.Thehumanubiquitinpromoterdrivingreportergeneexpressionisthestrongestmammalianendogenouspromotersthatweprovideformanycelltypes.TheIRESthatisusedtodriveluciferaseexpressionfromtheCMVpromoterisconsiderablyweakerthantheCMVpromoterandtypicallyyields15-20foldlowerproteinlevels.
